Genomic binding and transcription control by Elf1 in primary myeloid cells

The role of the transcription factor Elf1 was studied in murine myeloid precursor cells. For this purpose HSPCs were immortalized by HoxA9 and subsequently Elf1 levels were manipulated by shRNA and overexpression of HA-tagged Elf1 or degradable Elf1-FKBP. Genome wide Elf1 binding sites were determined by ChIP and genes under control of Elf1 were identified by RNA-seq after degradation of Elf1-FKBP. In addition upstream regulation of Elf1 by MLL fusions and HoxA9 was confirmed by ChIP.
