In order to investiage the morphogenetic control of early tube development in the Drosophila melanogaster embryo we performed single-cell-RNAseq-analysis of FACS-isolated early salivary gland placodal cells (stage 10-12), making up less than 0.4% of cells within the embryo to look for early tubulogeneic signatures. In order to identify salivary gland specific signals from epidermal signatures we also performed single-cell-RNAseq-analysis on FACS-isolated epidermal cells in stage 10-12 Drosophila melanogaster embryos for the purpose of differential expression anaylsis. Overall design: Two batches of salivary gland (SrcGFP) and one batch of epidermal tagged cells (ArmYFP) were obtained and isolated by flow cytometry from Drosophila embryos stage 10-12, and analysed by single cell sequencing.
