Comparative genomic fitness of saturated Tn-seq libraries of <i>Brucella abortus</i> S19 in stress conditions

<i>Brucella abortus</i> is a globally significant zoonotic pathogen. To explore functional relevance of its genes, a high-density transposon sequencing (TnSeq) library of the attenuated <i>B. abortus</i> S19 strain was exposed to extended periods of cell-wall targeting antibiotics or antimicrobial peptide-induced stress, or shocked with a potent acidic or oxidative stress. After condition exposures, transposon (Tn) mutants were collected and DNA was enriched for the transposon before sequencing. TnDivA was used for analyzing these highly saturated transposon libraries for Tn richness in the <i>B. abortus</i> S19 genome. Here are the sequencing results and analysis of the Tn insertion frequencies, Tn effective densities, and subsequent statistical analyses to identify conditionally essential and influential genes.
