Defective elastin induces aortic smooth muscle cell hyperproliferation. As elastin is expressed from embryonic day 14 in the murine aorta, we utilized bulk RNA-seq to reveal molecular differences between Eln(+/+) and Eln(-/-) aortic smooth muscle cells at embryonic day 15.5. Overall design: Acta2-GFP, Eln(+/-) mice were bred with each other and embryos were collected at embryonic day 15.5. Aorta was isolated from Acta2-GFP embryos carrying Eln(+/+) or Eln(-/-). After digestion with collagenase enzyme mixtures, GFP+, CD31-, DAPI- smooth muscle cells were sorted by FACS and incubated with RNA lysis buffer. Extracted RNA was subjected to bulk RNA-seq.
