15.5K is a RNA binding protein involved in the Minor Spliceosome. Here, we used shRNAs to knock down 15.5K in HEK293T cells to analysis the function of 15.5K in ncRNA expression. Total RNA from negative control HEK293T (shNC) cells and two 15.5K-knockdown (sh15.5K-1 and sh15.5K-2) cells were ligated to RNA adaptors, followed by rRNA and snRNA depletion. After reverse transcription and library amplification, inserts between 20-500bp were size selected and subjected to Illumina sequencing.
