Uterine-specific SIRT1 deficiency confers premature uterine aging and impairs invasion and spacing of blastocyst and stromal cell decidualization in mice.

We demonstrate that Sirtuin 1 (SIRT1) is an indispensable age-related regulator of invasion and spacing of blastocysts, as well as decidualization of stromal cells. Here, we report the sequencing of the uterine transcriptome of control and conditional knock-out of the Sirtuin 1 (SIRT1) at day 3.5 of murine pregnancy. Mice with uterine-specific ablation of Sirt1 (Sirt1d/d) and littermate controls (Sirt1f/f) were mated to male mice of proven fertility. Upon the presence of the post-coital plug (Gestational day 0.5, GD0.5), mice were sacrificed 3 days later (GD3.5) and uterine horns (embryo were flushed out) were harvested and frozen. RNA was isolated from the uterus. Six biological samples per genotype were sequenced for differential gene expression analysis.
本研究证实,沉默信息调节因子1(Sirtuin 1,SIRT1)是调控囊胚侵袭、排布以及基质细胞蜕膜化的不可或缺的年龄相关调控因子。本研究报道了小鼠妊娠第3.5天时,对照组与Sirtuin 1(SIRT1)条件性敲除小鼠的子宫转录组测序结果。实验中,将子宫特异性敲除Sirt1的小鼠(Sirt1d/d)及其同窝对照小鼠(Sirt1f/f)与经证实具有生育能力的雄鼠进行合笼交配;当观察到交配栓时记为妊娠第0.5天(GD0.5),于3天后(GD3.5)处死小鼠,收集已冲出胚胎的子宫角并进行冻存;随后从子宫组织中提取RNA,每个基因型设置6个生物学重复样本进行测序,以开展差异基因表达分析。
