Embryonic callus induced from maize immature embryos is widely used in genetic transformation of maize. However, only the limited inbred lines possess the ability to efficiently induce embryonic callus, which has highly inhibited transgenic breeding and revelation of gene function in maize. Since the plant regeneration from maize tissue culture was firstly reported, little is known about how the maize embryonic callus was induced from the immature embryos even though great efforts have been made by generation to generation of researchers. To date, only some QTLs have been identified using different populations, and a few genes were reported to control embryonic callus formation. But the molecular mechanism underlying embryonic callus induction is still obscure, due to the enormous sequence variation among maize genomes. To accelerate such efforts and to understand the basis of the chromosomal context, we de novo sequenced and assembled of a high callus induction ratio inbred line A188 by PacBio long-read sequencing and Bionano optical map technologies, which allow for a more detailed genomic analysis.
