RNA signatures followed by immunization of multiantigenic adenovirus-based vaccine candidate against mycobacterium tuberculosis

We here report that recombinant multiantigenic adenovirus based vaccine candidate induces immunogenicity and protective effectiveness. RNA-seq transcriptome analysis on whole blood of vaccine canddate vaccinated mice collected pre-, 1 week post-, and 4 weeks postinfection revelaed differentially expressed genes associated with immune and inflammaotry responses, especially belonging to the Wnt signaling pathway. Our results suggest that immunization with vaccine candidate promotes strong boosting effects in BCG-primed mice, presenting itself as a new vaccine candidate. Overall design: To investigate the RNA signature induced by vaccine candidate, mice were vaccinated with BCG pasteur 1173P2 via the subcutaneous route at 5-6 weeks of age, and after 10 weeks, immunized twice with vaccine candidate at 4 week intervals. Three weeks after the final immunization, mice was challeged with Mycobacterium tuberculosis H37Rv strain using a Glas-col aerosol generator. To evalute RNA signature, whole blood of mice was collected at pre-challenge, 1 week post-challenge, 4 weeks post-challenge.
