Colorectal cancer aberrant repeat RNA expression can be therapeutically targeted with reverse transcriptase inhibitors. Overall design: The experiments performed were on colorectal cancer (CRC) cell lines in different conditions to characterize the effects of P53 status and therapy on CRC. Cell lines in vitro were grown in 3D tumorsphere conditions and treated with DMSO control or 3TC with biological replicates at Day 1 and Day 7 post-treatment for a total of 96 RNA-seq samples. Cell lines grown as xenografts in Nu/Nu mice were treated with PBS control, 3TC, AZA, or 3TC + AZA for a total of 51 tumors with RNA-seq. ChIP-seq was performed on cell lines in vitro comparing input and p53 immunoprecipitation (IP) for a total of 12 samples.
