Previous studies have used this model to show that PC-TP has a protective effect when challenged with a methionine and choline depleted diet, known to induce NAFLD. We performed transcription factor binding experiments and show that PPARs are dysregulated in the contect of PC-TP depletion We then performed gene expression profiling analysis using data obtained from RNA-seq of 3 Mice for WT and KO fed either normal chow or methionine and choline depleted chow. 既往研究借助该实验模型证实,PC-TP在小鼠暴露于可诱导非酒精性脂肪性肝病(Non-Alcoholic Fatty Liver Disease, NAFLD)的蛋氨酸胆碱缺乏(Methionine and Choline Depleted, MCD)饮食时,具有保护作用。我们开展了转录因子结合实验,结果显示在PC-TP缺失的背景下,过氧化物酶体增殖物激活受体(Peroxisome Proliferator-Activated Receptors, PPARs)表达失调。随后,我们利用3只野生型(Wild Type, WT)与3只基因敲除型(Knock Out, KO)小鼠的RNA测序(RNA-seq)数据开展基因表达谱分析,这些小鼠分别饲喂普通饲料或MCD饲料。本研究针对饲喂MCD饲料或普通饲料的野生型与基因敲除型小鼠肝脏的RNA测序数据,完成了对比基因表达谱分析。
