To ask whether perturbations on global O-GlcNAc levels could alter embryonic stem cell identity and differentiation processes, we explored the gene expression of mouse embryonic stem cells (mESCs) and retinoic acid (RA)-inducted differentiated cells with or without OGT inhibition by per-acetylated 2-acetamido-2-deoxy-5-thio-D-glucopyranose (Ac45SGlcNAc, 5S). Overall design: We performed gene expression profiling analysis using data obtained from RNA-seq of mESCs and retinoic acid (RA)-inducted differentiated cells with or without OGT inhibition by Ac45SGlcNAc. For OGT inhibition, mESCs were treated with 25 μM Ac45SGlcNAc for 48 h. For inducing mESC differentiation, the cells pre-treated with DMSO or Ac45SGlcNAc were then incubated with 1 μM RA for 48 h. N_NC: normal mouse embryonic stem cells, N_5S: 5S treated mouse embryonic stem cells, DIF_NC: normal differentiated cells, DIF_5S: 5S treated differentiated cells.
