Foxa1 mutants induce distinct transcriptomic and DNA accessibilities, prominent aberrant epithelial lineage gene expression, and enhance prostate tumorigenicity in vivo Overall design: Murine organoids established from adult mouse prostate were transduced with lentivirus for pCW-EV and pCW-Foxa1 (WT and mutant cDNA). Following sorting and 3D culturing organoids, single nuclei were extracted from mouse prostate organoids. Joint scRNA- and scATAC libraries were prepared using the 10x Genomics platform.
