Quantitative PCR (qPCR) results showing mRNA expression levels of leptin receptor isoforms (LepR1–LepR5) in adrenal tumor and control tissue samples. RNA was isolated from adrenal tissues, reverse-transcribed to cDNA, and analyzed using SYBR Green qPCR on a Bio-Rad CFX96 system. B2M served as the reference gene. Each column represents: B2M.Cq – reference gene Cq; lep1.Cq, lep2.Cq, lep3.Cq, lep5.Cq – Cq values for leptin receptor transcript variants; lep1_dd–lep5_dd – ΔCq (difference between target and B2M); ddCt_lep1–ddCt_lep5 – ΔΔCq relative to control mean; and 2^-ddCt_lep1–2^-ddCt_lep5 – normalized relative expression (fold change). Empty cells indicate undetectable expression. Primer pairs used in the qPCR correspond to specific leptin receptor transcript variants: lep1 → transcript variant 1 (LepR1), lep2 → variants 2 and 4 (LepR2&LepR4), lep3 → variants 3 and 6 (LepR3&LepR6), lep5 → variant 5 (LepR5). Cq values represent the number of amplification cycles required to reach the fluorescence threshold and are inversely related to expression level. Relative expression was calculated using the 2^-ΔΔCq method.
