Understanding how endogenous ROS levels are controlled during normal aerobic growth remains uncharacterized with Mtb. Identifying regulators of eROS is likely to reveal new mechanisms of redox homeostasis that can be explored for therapeutic benefits. Here, we combined the Mrx1-roGFP2 redox biosensor with transposon mutagenesis and flow-sorting to identify regulators of redox metabolism in Mtb at the genomic level. We identified a set of 365 genes (redoxosome) responsible for maintaining non-toxic levels of eROS in aerobically grown Mtb.
