BCL11A interacts with and recruits the chromatin remodeller, CHD8 and this interaction is essential for mediating the oncogenic activity of BCL11A. Overall design: BCL11A shRNA sequences ((TRCN0000033449) mCHD8 (TRCN0000217316) were obtained from TRC consortium and cloned into a piggyBac transposon vector (PB-H1-shRNA-GFP) as described previously (Khaled 2015). 4T1 were transfected with 4.0ug of respective vector or control (scramble) using Lipofectamine LTX (Invitrogen). Cells were treated with G418 (400 ug/mL) (Gibco) for 5 days after which GFP+ve cell were sorted and cultured. ATACseq assay was performed on cultured cells as below
