Hepatic arterial infusion chemotherapy with FOLFOX regimen which contains oxaliplatin has been demonstrated to provide survival benefit, but chemoresistance substantially reduces response rates and clinical efficacy. To systemati-cally identify strategies that increase oxaliplatin sensitivity, we performed genome-scale negative-selection CRISPR-Cas9 screens in two hepatocellular carcinoma cell lines. DNA damage response pathways emerged as conserved sensitizers to oxaliplatin.
