Tissue fibrosis is a common pathological outcome of chronic disease that markedly impairs organ function leading to morbidity and mortality. In the lung, idiopathic pulmonary fibrosis (IPF) is an insidious and fatal interstitial lung disease associated with declining pulmonary function. Single cell RNA sequencing was used to map epithelial cell types of the normal human airway and alveolaor as well as IPF explant tissue. Overall design: Single cell RNA-seq using 10x chromium system on isolated epithelial cells (DAPI-,CD45-,CD31-,CD326+) from donor , and IPF patients lung explant tissue. Here we present data from 9 donor samples, 5 out of this 9 donors, we collect epithelial cells from tracheal, dissected proximal airway as well as distal tissue to represent all epithelial cell types of donor lung. The other 5 donor samples, we only collected distal lung tissue based on availability of tissue. For IPF explant tissue, we also present 11 ipf samples of fibrotic region of IPF lungs. Note, for dissection of airway tree, there is inevitable some distal connective tissue, that's why AT2 and AT1 cells presents in the proximal epithelial fraction.
