Low EIF4G2 protein expression was shown to have a poor OS of endometrial patients. Control and EIF4G2KD HEC-1A cells were sorted using CD133 merker to CD133+ and CD133- populations. These populations were taken to RNAseq and MS analysis to explore the mechanism. Overall design: HEC-1A endomertial cell line was KD with shRNA for EIF4G2 (DAP5) or shGFP (shNT) as control. The control and EIF4G2KD cells were subjected to sorting according to CD133 into CD133+ (p) and CD133- populations. The four populations were sorted as N=4 in independent experiments and save in the appropriate buffer according to the manufacturers protocol (Dynabeads™ mRNA DIRECT™ Purification Kit (Invitrogen)). mRNA was isolated and subjected to MARSseq2.0 protocol.
