In this preliminary proof-of-concept study, we demonstrate for the first time that intestinal stem cells derived from pre-term infant gut organoids can be successfully bioprinted using a Reactive Jet Impingement (ReJI) technique and maintained in a defined hydrogel. Stem cells were isolated from established pre-term ileal organoids, embedded in a collagen–alginate–fibrin (CAF) hydrogel, and subjected to ReJI-based printing. Post-printing viability was confirmed via Live/Dead and PrestoBlue assays, while confocal imaging verified retention of stem and differentiated epithelial phenotypes. Despite mechanical stresses associated with printing, cells remained viable and exhibited lineage differentiation capacity comparable to Matrigel controls.
