Investigating Ngn3-expressing cells in the adult mouse pancreas

The beta cell mass of the adult pancreas is thought to be maintained through low-level proliferation. Ductal cells have been proposed as an alternative source of beta cells in vivo, but this remains controversial. By performing detailed lineage tracing analysis, 3D imaging and single cell RNA sequencing we investigated if ductal cells contribute to the beta cell mouse in the adult mouse. Islets isolated from 6 mice of the genotype Ngn3-CreERT: R26-CAG-TdTomato (10 days post-tamoxifen) were dissociated to single cells and analysed using 10X chromium system.
