We report the application of RNA sequencing technology for high-throughput expression profiling in human Induced Pluripotent Stem cells differentiated toward skeletal muscle and motor neurons lineages. Transcriptomic analysis was performed at different time points during the differentiation process Transcriptome of biological duplicates at 3 key time point of the differentiation process. Briefly, hIPSCs derived from two different primary fibroblasts cell lines were processed throughout a differentiation protocol allowing the production of stable and mature muscle fibers along with motorneurones structures. A total of 6 Differenciation procedures were carried out with the same hIPSC clones for a total of 3 time point in biological duplicate. Results were analyzed to reveal transcritpome dynamics. RNA Seq performed at different time point of a differentiation procedure; Each differentiation corresponds to one RNA Seq, as one cannot divide cells or split samples during this process. Thus, 6 differentiations correspond to 2 differentiations stopped at D8, 2 stopped at D17 and 2 stopped at D30. Differentiations are performed from human Induced Pluripotent stem cells (hIPCs) derived from the primary fibroblast.
