Duck tissue metagenome

For viral metagenomic analysis, 192 samples from 36 death ducks were randomly collected from the same farm in Shandong Province, China, 2021. The 192 samples were randomly pooled into eleven supernatant pools, each including 16-18 samples. Each supernatant pool was filtered with a 0.45um filter to remove particles the size of bacterial and eukaryotic cells and the filtrates enriched in viral particles were treated with DNase and RNase to digest unprotected nucleic acid. The remaining total viral RNA and DNA were extracted by using the QIAamp viral RNA Minikit according to the protocol. Eleven libraries were then constructed using Nextera XT DNA Sample Preparation Kit (Illumina) and sequenced using the MiSeq Illumina platform with 250 bases paired ends with dual barcoding for each pool.
