Collectively, our study reveals the role of Olig2 in hindering the in vivo AtN conversion and idendifies an inducible barrier to neuronal reprogramming. Overall design: WT mouse cortex was injected with AAV9 hGFAP-Ngn2-tdTomato in the presence of scramble shRNA or Olig2 shRNA. Virus-infected brain tissue was collected 18, 20, 22 days post injection (n = 2 mice for each time point), and dissociated into single-cell suspension, which was loaded to microfluidic chip, followed by scRNA-seq processing.
