Analysis of drought stress responsive SSH library during flowering stage in Pennisetum glaucum cv 841B. Pg cv. 841B SSH DS F library

Seeds of drough tolerant Pennisetum glaucum cv 841B were grown under in vitro conditions. Plants, at flowering stage were subjected to drought stress by water withdrawl at different time interval i.e. 10 days, 15 days, 20 days, 25 days. Total RNA was isolated from leaf tissue at different time intervals and cDNA was prepared from the pooled RNA. EST library was prepared through SSH (Suppression Subtractive Hybridization) technique. Secondary PCR products were ligated to pGEM-T easy vector and transformed into E. coli strain DH5a. The clones were sequenced using T7 primer. A total of 889 ESTs were generated
