从花白头发的人头皮样本中分离并高度纯化了成对的黑发和白发毛囊样本,进行了单细胞转录组测序分析。基于对这些数据的分析和额外临床样本验证,绘制人类头皮毛囊上皮组织及其干细胞的单细胞转录组图谱,并揭示了其中多种前所未知的细胞分化途径和毛囊干细胞标志物,为今后的人头皮毛囊研究提供了基础蓝图。随后,通过黑白毛囊数据对比与额外临床样本验证,我们发现毛发前体细胞而非毛囊干细胞的枯竭是人头皮毛囊衰老白化的早期机制。结合小鼠模型研究,我们发现P53通路激活是导致这种毛发前体细胞枯竭的重要机制,而在皮肤表面涂抹P53抑制剂能够有效拮抗包括白发在内的毛囊衰老早期症状,从而指出了一种极具临床潜力的毛囊衰老白化拮抗途径。
Paired black and white hair follicle samples were isolated and highly purified from human scalp specimens obtained from individuals with mixed gray/white and black hair, followed by single-cell transcriptome sequencing analysis. Based on the analysis of these data and validation with additional clinical samples, we constructed a single-cell transcriptomic atlas of human scalp follicular epithelium and its stem cells, uncovered multiple previously unknown cell differentiation pathways and follicular stem cell markers, and provided a foundational blueprint for future research on human scalp hair follicles. Subsequently, through comparative analysis of transcriptomic data from black and white hair follicles and validation with additional clinical samples, we identified that the depletion of hair follicle progenitor cells (rather than follicular stem cells) represents the early mechanism underlying human scalp hair follicle aging and graying. Combined with studies in mouse models, we found that activation of the P53 signaling pathway is a critical driver of this depletion of hair follicle progenitor cells. Topical application of P53 inhibitors on the skin effectively antagonizes early symptoms of hair follicle aging including graying, thereby identifying a clinically promising strategy for antagonizing hair follicle aging and graying.
