Cleavage Under Targets and Tagmentation（CUT&Tag）analysis of ovarian cancer cells treated by Paeonol

Cells were harvested and aliquoted (500,000 cells). Cell nucleus was prepared and resuspended. Incubation of the cells was performed with ACSS2 and IgG primary antibodies throughout the night at 4℃. The cells were centrifuged and washed using Dig-Wash buffer. A dilution of the pA-Tn5 adapter complex was introduced into the cells, followed by incubation at RT for 1 h. Subsequently, the cells were subjected to incubation in Tagmentation buffer for 1 hour at 37℃, and the STOP buffer was eventually added. The DNA library was purified and amplified for sequencing, and CUT&Tag data analyses were performed. CUT&Tag analysis was performed on ovarian cancer cells (SKOV3 cells) after being treated by 2DG for 24 hours
