The fungal composition of oil treated wood surfaces, that contained the specific mould stain formation called biofinish, has been analysed. A culture-based approach and two culture independent methods were used. In the culture-based approach biomass was obtained from the surface of oil treated wood samples exposed outdoors at different locations. Culturing the biomass on agar plates showed the presence of a few predominant types of colonies. The genus Aureobasidium and an unclassified coelomycete were most frequently detected as predominant constituents of biofinishes. Additionally, a culture independent approach was carried out, based on ITS specific PCR amplification followed by cloning and sequencing. In order to get an in depth overview of the culture independent composition of wood samples exposed in the Netherlands, ITS1-specific Miseq amplicon sequencing was applied on various combinations of wood species and (oil) treatments.
