Dataset for the Intronic Signaling Pathways System (ISPS): Structural and Interaction-Based Analysis of Intronic RNA Across Inflammatory Genes

This dataset contains processed structural and interaction-based metrics supporting the study "Intronic RNA as organized regulatory platforms: a quantitative framework for the Intronic Signaling Pathways System (ISPS)". The dataset includes intron-level analyses across representative inflammatory genes (CRP, TNF, IL6, IL8, and IL1B), integrating RNA secondary structure prediction, transcription factor binding site (TFBS) identification, and RNA-binding protein (RBP) interaction profiling. Intronic sequences were computationally reconstructed into lariat-like configurations and analyzed using RNAfold for secondary structure prediction. TFBS were identified using FIMO with JASPAR databases, and RBP interactions were derived from ATtRACT, RBPDB, POSTAR3, and CISBP-RNA. Structural and interaction features were integrated to compute ISPS-derived metrics, including the Intronic Complexity Index (IICI). The dataset is organized by gene, with each folder containing processed intron-level metrics and interaction data. For IL8, RBP interaction data are provided as platform-specific outputs used to derive integrated interaction profiles described in the manuscript. These data support the identification of distinct intronic regulatory architectures, including monotonic, hierarchical, biphasic, and multimodal ISPS gradient profiles. All files are provided in processed formats to facilitate reproducibility and downstream analysis.
