Single cell RNA sequencing on rat fetal hypothalamus in a context of nutritional protein restriction

Female rats were fed a control (C: 20% protein) or protein restricted (PR: 8% protein) for 4 weeks before mating. From the first day of gestation, PR rats turn to a 4% protein diet. Fetuses were sacrificed on gestational day 15 (G15), hypothalami were dissected and mechanically dissociated into individual cells. Cells were allowed to proliferate in standard culture conditions for 48 hours. Cells were dissociated and passed on a facs ARIA-III prior to loading onto the CHROMIUM Chip (10X Genomics).The aim was to assess the impact of protein deficiency on the various hypothalamic precursor cells in the fetal stage.
