RNA-seq of leaf and root tissues from T. kiharae and its tetraploid (T. timopheevii) and diploid (Ae. tauschii) parents

Plants of T. kiharae and its tetraploid (T. timopheevii) and diploid (Ae. tauschii) parents were grown in the same growth chamber under a 16-h light/8-h dark photoperiod with a light intensity of 225 μmol m−2 s−1 and a 25 °C/18 °C (light/dark) cycle. Three biological replicates were collected from the second leaves and roots of third-leaf-stage T. kiharae, T. timopheevii, and Ae. tauschii seedlings and stored at −80 °C for total RNA extraction. Total RNA was isolated from the leaf and root tissues using TRIzol reagent (Invitrogen) according to the manufacturer’s instructions. Then, the RNA-seq were performed.
