Plants of T. kiharae and its tetraploid (T. timopheevii) and diploid (Ae. tauschii) parents were grown in the same growth chamber under a 16-h light/8-h dark photoperiod with a light intensity of 225 μmol m−2 s−1 and a 25 °C/18 °C (light/dark) cycle. Three biological replicates were collected from the second leaves and roots of third-leaf-stage T. kiharae, T. timopheevii, and Ae. tauschii seedlings and stored at −80 °C for total RNA extraction. Total RNA was isolated from the leaf and root tissues using TRIzol reagent (Invitrogen) according to the manufacturer’s instructions. Then, the RNA-seq were performed. All transcriptome data generated in this study are also available at the National Center of Biotechnology Information under accession number PRJNA1221302 (https://www.ncbi.nlm.nih.gov/sra/).
