In mammals, the first cell fate decision occurs in the 2-cell (2C) stage, resulting from the totipotent zygote into two different lineages: the inner cell mass (ICM) and trophectoderm (TE). Cells from ICM can be maintained under the conventional LIF/Serum culture condition, containing a small percentage of cells that share features with the 2-cell embryo. These cells mirrored the in vivo totipotency with expanding developmental potential by yielding both embryonic and extraembryonic lineages. Mounting evidence has suggested Dux (Dux in mice, DUX4 in humans) is a master regulator of totipotency in mouse 1-4. However, almost all the research focuss on the transcription control of Dux, and lacks evidence showing how DUX protein regulates MERVL/2C genes expression on the epigenetic level.
