Analyzing differences in transcriptome profile of Rv?sigA (SigA conditional knockdown strain) in presence of anhydrotetracycline and transcriptome profile of Rv?sigB (SigB knockout strain) compared to control. Overall design: Rv?sigA was inoculated at 0.05 OD, in presence or absence of 1mM/ml anhydrotetracycline. Post 3 days total RNA was islolated and RNA seq was performed. Rv?sigB was inoculated at 0.05 OD and grown to 0.8 OD, after which RNA was isolated and RNA sequencing was carried out.
