River water and treated wastewater samples from the Kansai region of Japan were analyzed using amplicon sequencing targeting full-length 16S rRNA genes. For validation, a standard mock community sample (NBRC Microbial DNA Cocktail, DNA-Mock-001; Nite Biological Resource Center) was also sequenced. The study aimed to simultaneously detect bacterial pathogens at the species level and characterize pathogen communities in drinking water sources. Sequencing was performed using the MinION Mk1B (Oxford Nanopore Technologies), with unique molecular identifiers introduced during library preparation to reduce sequencing errors.
