In this study, we investigated the mechanisms of neuroprotection of factors secreted by glial progenitro cells. We earlier obtainedn data on their neuroprotective action on the model of glutamate excitotoxicity, as well as proteomic analysis. The model was reproduced on cortical neuron cultures from rat pups (P0, 10 DIV). Before the model reproducing, the samples were added. Next day a solution containing 100 µM glutamate was added to the cultures for 1 hour. The negative control was intact cultures. The addition of 100 µM memantine was a positive control. Three repeats were performed for each group and both controls. The comparison groups were: extracellular vesicles, proteins of the 3-100 kDa fraction and total secretome. Total RNA was collected 4 hours after model reproducing for transcriptome analysis to identify signaling pathways activated in cells by druges under induced glutamate toxicity. Overall design: A model of glutamate excitotoxicity was reproduced on neuroglial culture (10 DIV). A day before the addition of 100 µM glutamate, examples of extracellular vesicles, proteins of the 3-100 kDa fraction, and total secretome (3-infinity) were added to the cells. 100 µM memantine was used as a positive control. Total RNA was obtained 4 hours after reproducing of the glutamate excitotoxicity model.
