The SNORD115 snoRNA has been proposed to regulate A-to-I editing of the 5-HT2C pre-mRNA. We prepared a mouse strain where the SNORD115 locus is deleted, as well as a strain where both SNORD115 and the neighboring SNORD116 locus are deleted. Because these snoRNA loci are submitted to genomic imprinting (only the paternal allele is expressed), we compared WT specimens (which express SNORD115 and SNORD116) to paternally-inherited deletions (which do not express SNORD115, or: which do not express SNORD115 nor SNORD116, depending on the sample). For each dissected brain area in each replicate, the edited segment of 5-HT2C mRNA was amplified by RT-PCR and deep-sequenced.
