Representation of the plasmid library for transporter disruption of S. cerevisiae

This study is to verify the representation of the plasmid library for transporter disruption of S. cerevisiae. There are five separate sub-libraries Sctrans-PP-Grp1, Sctrans-PP-Grp2, Sctrans-PP-Grp3, Sctrans-PP-Grp4 and Sctrans-PP-Grp5, and a Sctrans-PP, which was generated by mixing the sub-libraries in the equal molar for all the individual plasmids.All of the gRNA-donor plasmids contain a similar construct with different sequences of the gRNA and donor. The gRNA-donor fragments can be PCR amplified with a primer sets targeting the consensus region, and fragments from different libraries can be pooled for the sequencing with Miseq (pair-end 300 bp reads).
