We report that the Pygopus (Pygo) 2 chromatin effector is associated with the regulatory elements and utilized for the expression of a set of MYC-dependent genes necessary for cell division. Global chromatin association studies revealed enrichment of Pygo2 at transcriptional start sites in which approximately 60% of Pygo2-associated genes were shared with MYC oncogene. Pygo2 and MYC were co-enriched at promoters/enhancers along with the histone mark H3K27Ac at critical MYC target genes involved in mitotic cell division. Pygo2 interacted with MYC and was required for MYC-dependent transcription and cell growth in a MYC-driven prostate tumor cell model. Our evidence supports a role for Pygo2 in a MYC-dependent transcriptional program that directs formation of the mitotic apparatus.
