CD12 is a novel bacterial phylum documented recently. Due to incompleteness of its genomes, metabolic style and ecological importance remain unclear. In this study, we identified a lineage of CD12 from a saline sediment sample near a cold seep in the Red Sea. Its affiliation to CD12 was supported by phylogenic relationships of 16S rRNA sequences. A collection of the representative 16S rRNA sequences of CD12 in public database showed a wide distribution in deep-sea sediments. Using metagenomic binning method, we obtained the draft genome of the CD12 lineage. Core metabolisms are centralized with glycolysis and pyruvate fermentation to produce acetyl-CoA. The genome also possessed transporter genes for a variety of sugars, indicating the capacity of the CD2 lineage in consumption of the hydrocarbons from environments. CO2 fixation could be conducted through Wood-Ljungdahl pathway by this lineage. The resulting acetyl-CoA might be then converted to acetate. The draft genome of the acetogenic bacteria allowed us to reveal the saccharolytic and fermentative mode of metabolisms, coupled with autotrophic CO2 assimilation processes. Thus, candidate phylum CD12 is probably a heterotrophic supplier of acetate in deep-sea sediments.
