We develop a non-invasive method for comprehensive characterization of the extracellular RNAs (exRNA) in a single droplet of spent media that was used to culture human in vitro fertilization (IVF) embryos. We generate and provide as a resource the temporal extracellular transcriptome atlas of human preimplantation development (TETA). TETA consists of 245 exRNA sequencing datasets, corresponding to 245 collections of spent media at five developmental stages. The spent media in their original culture dishes were obtained immediately after embryo transfer to fresh media. The fertility center follows a 7-day standard procedure. A total of 261 spent media droplets were collected at four time points, 18h, 68h, 120h, and 160h. Additionally, a set of unfertilized oocytes were cultured for up to 68 hours. A total of 34 droplets of oocyte spent media were collected. The spent media were carefully aspirated out from the original culture dishes and centrifuged at 500 ×g for 3 min under 4°C. The supernatant was aspirated and centrifuged again. A 7 μL aliquot of the supernatant of the 2nd round of centrifugation was used as the input to SILVER-seq. NOTE FROM SUBMITTER: Due to sensitivity of the raw data in our project, we've decided to upload only the processed data. We will deposit the raw data to dbGaP.
