Developmental processes mediated through changes in gene expression from the nuclear genome are regulated at the level of local chromatin structure. Increased accessibility of gene-proximal DNA to diffusible nucleases has previously been shown to correlate with increased gene expression defined as increased steady-state levels of polyadenylated mRNAs relative to control or other tissue types. We applied a Micrococcal Nuclease Differential Nuclease Sensitivity assay coupled to next generation sequencing (MNase DNS-seq) to profile genome-wide chromatin accessibility in several developmentally distinct tissues of Zea mays. Differential sensitivity between Ear shoot, Root Tip, Coleoptilar Node, and 15 day after pollination Endosperm identifies differentially utilized regulatory loci in these disparate tissues.
