Bulk RNA-seq reveals effects of tamoxifen on endothelial gene expression

Transgenic line combination with tamoxifen induction is widely used for biological study. Considering the possibility that tamoxifen induction may alter gene expression, we performed this study to elucidate the potential effect of tamoxifen gavage on the gene expression profile of endothelial cells. Cdh5-CreERT2(PAC);Rosa26fsTRAP transgenic mice were divided into 2 groups: with tamoxifen gavage group and withou tamoxifen gavage group.Tamoxifen gavage was conducted at E12.5 for pregnant mice, then E15.5 embryos were harvest to perform endothelial cells (ECs) sorting with FACS. ECs purified from different samples were stored at -80℃ and then processed at the same time to produce sequencing libraries. rRNAs were deleted with NEBNext rRNA Depletion Kit (V2) and libraries were made with NEBNext Ultra II RNA Library Prep Kit.
