The expression of Eukaryotic translation initiation factor 2 subunit 3 (EIF2S3/eIF2γ) in patients with non-small cell lung cancer and colorectal cancer is lower than that in healthy people, but its functions and mechanisms are still unclear, and the study of EIF2S3 in adult leukemias has not been reported. The expression of EIF2S3 in acute myeloid leukemia (AML) patients was lower than that in patients with complete remission (CR) and significantly associated with clinicopathological parameters and prognosis. EIF2S3 overexpression inhibited cell proliferation, blockaded the progression of cell cycle from G0/1 to S phase, inhibited the tumorigenicity and decreased the phosphorylation level of ERK protein in AML cells. In conclusion, EIF2S3 could be expected to be a novel therapeutic target in AML. We used Retro-X Tet-Off Advanced doxycycline (DOX) inducible expression system to generate stable EIF2S3 overexpressed cell (HL60-EIF2S3). The targeted gene, EIF2S3, was suppressed in the presence of DOX.
