BCL11A interacts with and recruits the chromatin remodeller, CHD8 and this interaction is essential for mediating the oncogenic activity of BCL11A. BCL11A shRNA sequences ((TRCN0000033449) mCHD8 (TRCN0000217316) were obtained from TRC consortium and cloned into a piggyBac transposon vector (PB-H1-shRNA-GFP) as described previously (Khaled 2015). 4T1 were transfected with 4.0ug of respective vector or control (scramble) using Lipofectamine LTX (Invitrogen). Cells were treated with G418 (400 ug/mL) (Gibco) for 5 days after which GFP+ve cell were sorted and cultured. ATACseq assay was performed on cultured cells as below
