Human TSLP administration promotes homeostatic maturation of human cDC2 in humanized mice

To test whether TSLP triggers homeostatic maturation of human cDC2s in vivo, we administrated human TSLP to humanized mice and performed scRNA-seq. We found that TSLP administration increased the proportion of homeostatically matured DC (DC2hm) subtype in cDCs. Overall design: Humanized mice (male or female, 7 weeks old). At Day 0, human TSLP (1mg/kg) was injected intraperitoneally, and then every 2 days. The control group mice were injected intraperitoneally with the same volume of PBS. On the 6th day, the mouse spleen was taken out and ground into a single-cell suspension. hCD3+, hCD19+ and hCD14+ cells were firstly deleted from splenocytes using MACS, and then purified hCD45+cells from the effluent were collected for scRNA-seq or flow cytometry.
