Gene Expressoin Profile in the Liver of Male WT and BGN_KO Mice at 13 Weeks Old

To investigate the effects of gene Bgn on affecting brain dysfunction and metabolic disorders by profiling the transcripome in the liver of male Bgn_KO and wild-type (WT, litter mate) mice at 13 weeks old. All mice (male WT_liver (n=4) and male BGN_KO_liver (n=4)) were maintained in a temperature-controlled room with a 12-hour light and 12-hour dark cycle. Diet (Pico Lab 5053 rodent diet) and water were provided ad libitum. At 13 weeks old, mice were sacrificed. Hypothalamus and liver were dissected out, flash frozen, and stored at -80°C for later usage. All experiments were performed in accordance with the United States National Institutes of Health Guide for the Care and Use of Laboratory Animals and were approved by the University of California at Los Angeles Chancellor's Animal Research Committee.

本研究旨在通过对13周龄雄性Bgn基因敲除(Bgn_KO)与野生型(WT,同窝对照)小鼠肝脏的转录组(transcriptome)进行谱分析,探究Bgn基因对脑功能障碍与代谢紊乱的影响。所有实验小鼠(雄性野生型肝脏组n=4、雄性Bgn敲除肝脏组n=4)均饲养于温度可控的环境中,光照周期为12小时明、12小时暗。实验采用Pico Lab 5053啮齿类饲料与饮用水自由供应的饲养方式。待小鼠生长至13周龄时实施安乐死,摘取其下丘脑与肝脏组织,经快速冷冻后保存于-80℃环境中以待后续实验。所有实验均严格遵循美国国立卫生研究院《实验动物护理与使用指南》开展,并经加州大学洛杉矶分校校长办公室动物研究委员会批准。
