CAR T Cell Sytotoxic Supernatant Stimulates Monocytes to Drive Cytokine Storm

Chimeric antigen receptor (CAR) T cell-therapy is substantially constrained by cytokine release syndrome (CRS). The precise mechanisms underlying CAR T cell-therapy-associated CRS remain elusive. Here, we show that CAR T cell-induced tumor cytotoxicity activates monocytes to release IL-1ß, IL-6, and TNF-a, key inflammatory cytokines driving CRS, via monocyte contact-independent manner. Overall design: THP-1 cell line was stimulated with PBS, Lipopolysaccharides, Mock T+Raji or CART+Raji supernatant,then THP-1 cells were collected for RNAseq
