Single-Cell Analysis of Aging Mouse Dentate Gyrus

The dentate gyrus of the hippocampus is a brain region involved in learning, memory formation, and spatial coding. We performed single-cell RNA-sequencing of the dentate gyrus of young and old mice to identify the age-induced changes. Mouse dentate gyri were microdissected and single cells were isolated via enzymatic dissociation. Viable cells were collected via fluorescence-activated cell sorting and subjected to 10x Genomics single-cell RNA-sequencing. Libraries were sequenced with Illumina NovaSeq 6000 28x94 Flow Cell SP, paired-end.
海马体的齿状回(dentate gyrus)是参与学习、记忆形成与空间编码的脑区。本研究针对年轻与老年小鼠的齿状回开展单细胞RNA测序(single-cell RNA-sequencing),以识别年龄诱导的基因表达变化。实验中,小鼠齿状回组织经显微切割获取,并通过酶解解离法分离单细胞;随后通过荧光激活细胞分选收集活细胞,采用10x Genomics平台完成单细胞RNA测序。测序文库使用Illumina NovaSeq 6000 28x94 Flow Cell SP进行双端测序。
