资源简介:
This dataset contains the raw and processed experimental data supporting the manuscript “TRPC6-Mediated Ca²⁺ Influx Activates MAPK and NF-κB Signaling to Drive Pro-Inflammatory and Catabolic Responses in Human Intervertebral Disc Cells.” Data were generated using primary human intervertebral disc (IVD) cells following pharmacological activation of TRPC6 with the selective agonist Hyp9 under serum-free and serum-containing conditions.Experiments were conducted to assess calcium influx dynamics, cell viability and cytotoxicity, inflammatory and catabolic gene expression, protein secretion, and downstream signaling pathway activation. All assays were performed using standardized protocols as described in the Methods section of the associated manuscript. No patient-identifiable information is included.File Structure and ContentsOverview–Description
Summary of experimental design, treatment conditions, and dataset organization.1_TRPC6 Endogenous Expression
Basal TRPC6 gene expression data across human IVD cell donors.2_Calcium Flux + Kinetics
Real-time calcium influx measurements (ΔF/F₀), area under the curve (AUC), time-to-peak, and EC₅₀ analyses obtained using a Fura-2 QBT assay on a FlexStation 3 platform following acute Hyp9 stimulation.3_Cell Viability
PrestoBlue assay data assessing metabolic activity and viability following TRPC6 activation.4_Cell Cytotoxicity
LDH release measurements evaluating membrane integrity and cytotoxic effects of treatments.5_Gene Expression
qPCR data for inflammatory, catabolic, and neuro-angiogenic markers following TRPC6 activation.6_Protein Expression
ELISA-based quantification of secreted cytokines, growth factors, and matrix-degrading enzymes.7_Phosphorylation
Western blot densitometry data for MAPK and NF-κB pathway components following TRPC6 activation. This dataset enables transparency, reproducibility, and independent re-analysis of calcium-dependent inflammatory signaling mediated by TRPC6 and will be of value to researchers studying TRP channels, intervertebral disc degeneration, and discogenic chronic back pain.
