LINE1 and PRC2 control nucleolar organization and repression of the 8C-state in human ESCs [RNA-Seq_RSeT+DT H9_EZH2i]

In this study, we investigated PRC2 (polycomb repressive complex 2), the H3K27me3 modifier, in naive hES cells for its roles in regulating 8-cell (8C) stage genes expression. EZH2 inhibitor, UNC1999 and its inactive analog UNC2400 were used for the treatment. Overall design: Naive H9 hES cells were maintained RSeT. 24 hours after cell splitting, cells were maintained in the RSeT media added with 10 nM DZNep and 5nM TSA (RSeT+DT), in combination with UNC1999 and UNC2400 added at 2.5 µM final concentration for the treatment and controls. 48 hours later, cells were directly processed for RNA extraction and RNA-seq library prep.
