The purpose of this study was comparation of the bacterial community between each step of the pH shift culture method. Soil suspension was spread on the specialized cellulose film method (Aoyagi et al. 2022) containing Reasoner's 2A and Lennox LB mediums. After 48 h, bacterial community formed on the cellulose film was transferred to another petri dish containing diluted H2SO4 (pH 1.5) and incubated for 48 h. Then, cellulose films were transferred to the medium again and cultured for 24 h. DNA of bacterial community formed on the cellulose film were extracted and the 16S rRNA amplicon sequencing was performed using Miseq (Illumina, Inc., San Diego, CA, USA.).
